There is a simple way of doing it.
Suppose you want to change the default player for .mp3 files to Mplayer
(assuming that it has been installed).
Just click on an .mp3 file, select
"Open with | Open with Other Application ...".
Choose the desired program from the list (i.e. Mplayer Movie Player).
Mplayer starts playing the file, and gnome automatically changes the default
file association (to .mp3 <=> Mplayer) for future use.
