Adding additional RAM

After inserting RAM into my iMac, G4, is there anything else I must do for the OS to use the additional RAM?

And, what application do I use to see how much RAM is installed?

No, as long as the motherboard supports the amount of ram your installing and supports the speed then you should be fine.
