What do you want?

An installation on a USB stick? In that case you need to create partitions yourself and do watch out for where the bootloader gets installed.

Or do you want a live USB with persistence? In that case, just use unetbootin and you don't need partitions.

A live USB can be used to install Ubuntu on other computers too. Otherwise it's an installation on USB.

The Live USB runs on a computer. A 64-bit USB will only run on 64-bit compatible processors, but the OS on that computer has nothing to do with it.

For Ubuntu based distros you can also use the program "startup disk creator".

You can't do an md5 check since your usb stick is touched (you can edit the files, and some files are changed before put on the USB).
