Is there any way to load up multiple linux flavors (such as ubuntu and its variations, backtrack, dsl, desbian,ect..) on to a USB drive with persistance?

Some persistence methods are not compatible with others.
Using the grub2 MultiBootUSB method you can have Puppy and Ubuntu persistent.

well....is there some way to perhaps boot off a non-persistant usb OS then make some changes, then some how repackage the OS to a seperate usb or iso to reinstall it on the drive with modifications?

essentially i was hoping to have maybe 6-8 OS's on a usb and and while i don't need to constantly change the files, i wanted to make certain adjustments such as extentions, networking, programs, and audio settings(don't need blaring audio on bootup in class >.<).

Not sure if that url is helpful but ill try it out when i get home
