Could someone please point me to a guide on how to upgrade my 9.10 to 10.04 using the iso I downloaded and burned?
Or is it easer to just hit the upgrade button in the "update manager"?

I would say use the upgrade button. Be aware though, upgrading takes a long time.

I don't think that a correct upgrade from an ISO is possible. Why? Well, because upgrading your system is a process that, among other things updates the packages of the applications you have installed. That's why you should upgrade your system using the online repositories and not a DVD/CD.
If you want to upgrade the way I suggested, I recommend you to update your installed packages, first.
sudo aptitude update
sudo aptitude upgrade
sudo aptitude dist-upgrade
