I have a GTX 295, with two DVI ports. If I insert another nVidia card in my spare PCIe-slot, will I be able to use it to output to a thrid monitor, or will it simply go unused?

That depends on your system.
In the past I had an AGP-card and a PCI-card. One Ati, one nVidia.
Some computers let me stick both in and use both at the same time. Other computers only saw 1 out of 2 cards.
