I could not update to ClamTk 4.16

ClamTk 4.16 released in July 11 2009 and I can't update as its not in synaptic, how I will be able to update it ?

The only way to get a newer version is to compile from source, or wait until Karmic Koala comes out, as the version your are using will not be updated, the only thing you will get is bug fixes.

When the new release "Karmic Koala" will come ?

After the new release, "Karmic Koala" released will i be able to update current system without format and keep current software installed and services ?

You could just get the .deb and install that.
