This is not supported. Your best bet is to use a 64-bit emulator (like qemu) to build a simple virtual machine server that just runs prevu.
Building 32-bit packages in a 64-bit is equally not well supported. Your best bet there is to set up a 32-bit chroot using stock prevu inside, or have a prevu32 that sets up its prefix in /var/cache/prevu32 instead of /var/cache/prevu; then a simple matter of pbuilder init invocation hacking can build a 32-bit tarball.

Thanks, where can I find info how to set up such a 64bit emulation with qemu?

Re: Build (backport) an newer Ubuntu package to run on an older Ubuntu version
ISn't it possible to upload the backport (made by prevu) to launchpad to enable amd64?

Is there a fix for Prevu in Lucid? I've seen the bug report but no fix
