As the subject says, I accidentally deleted a registry key. What is the easiest way to restore it? I don't mind reinstalling Wine if I have to.

Well, if you don't mind re-installing then the easiest way is to just delete the hidden .wine folder in your home dir. Press ctrl-h in nautilus (the standard file manager) to be able to see hidden files.
When that's done, to generate a fresh wine setup, just issue:
winecfg
And you're good to go.

Sounds pretty simple. Will I have to reinstall whatever windows applications I had in Wine?

Yes. BTW, do you know which registry key you deleted?

you should be able to do this without deleting the .wine folder. you may have to delete the registry files, so they can be rebuilt when wine is reinstalled and reconfigured.
Edit: creating new registry files may mess up the functionality of previously installed windows programs, depending on the program, so you may as well remove the entire .wine folder.

I was installing Gecko and deleted this key:
HKEY_Current_User/Software/Wine/MSHTML
No more MSHTML :(

- In that case, before getting rid of .wine, try:
wine iexplore http://www.winehq.org
- And if that fails, copy & paste the next lines in an empty text file:
[HKEY_USERS\S-1-5-4\Software\Wine\MSHTML]
"GeckoUrl"="http://source.winehq.org/winegecko.php"
[HKEY_USERS\S-1-5-4\Software\Wine\MSHTML\0.1.0]
"GeckoPath"="C:\\windows\\gecko\\0.1.0\\wine_gecko"
Name it "last-hope.reg" :) and import it into the registry using:
wine regedit last-hope.reg
