You can take a look at ZenWalk, they have an easy tutorial to set it up to boot from a flash drive.

I actually think i gave ZenWalk a try but something involving my X-Server wouldn't boot it up.

is best: unlike the weirder customized flash Linuxes, it's straight Debian, but still gives install scripts for flash and hd, and unlike damn small, has very latest kernel and packages.

mcnlive is a live cd with a great community and a wizard to both create a liveusb and a loop file to save setting. it also has a remaster on the fly wizard which is great for basically making mcnlive to your exact specifications. I use mcnlive and absolutely love it, it is based on mandriva. I tried to run ubuntu from a live usb it ran fine and all that but it just didnt feel right whereas mcnlive i would feel comfortable using on a daily basis

And even better, if you have a gig of ram or more, you can use the copy to ram option and it runs quicker and saves your flashdrive all those 'writes' and frees it up to be used as a storeage device.

Same goes for the livecd , use the copy to ram option and you can remove the cd from the drive and use the drive normally.
